The Six Human Trials, by Route and Amount
A 2013 safety paper in the Journal of Endocrinology and Metabolism pulled together all six human trials of AOD 9604, run in Australia between 2001 and 2006. As that paper reports them, the trials gave:
| Trial | Amount | Route | Length | Who |
|---|---|---|---|---|
| METAOD001, phase 1 | 25 to 400 mcg per kg | Drip into a vein over 20 minutes | 3 single doses, a week apart | People: 15 healthy men, BMI 24 to 30 |
| METAOD002, phase 2a | 25, 50 and 100 mcg per kg | Drip into a vein over 20 minutes | Single doses, a week apart | People: 23 obese men, BMI 35 or more |
| METAOD003, phase 2a | 9, 27 and 54 mg | By mouth, capsules | Single doses, 2 weeks apart | People: 17 obese men, BMI 35 or more |
| METAOD004, phase 2a | 9, 27 or 54 mg a day | By mouth, capsules | 7 days | People: 36 obese men, BMI 30 or more |
| METAOD005, phase 2b | 1, 5, 10, 20 or 30 mg a day | By mouth, capsules | 12 weeks | People: 300 obese men and women, BMI 35 or more |
| METAOD006, phase 2b | 0.25, 0.5 or 1 mg a day | By mouth, tablets | 24 weeks | People: 502 obese adults, BMI 30 to 45 |
Disclaimer: these are the amounts six company trials gave, quoted so you can see what was tested. They are not a dose for you, and no major regulator has approved AOD 9604. A licensed clinician makes any dose decision.
Look at the route column. Two trials used one drip into a vein, dosed by body weight. Four used capsules or tablets swallowed in mg amounts. None gave AOD 9604 as a shot under the skin, so the daily amounts in mcg that forums post for that route were never what these trials tested.
Two trials were built to measure weight loss, yet the 2013 paper reports only safety results. PubMed lists no trial report for AOD 9604 at all, while the same search filter does find trials for other peptides. All six trials were paid for by the company that developed AOD 9604, and the paper's authors include people tied to that company.

Animal Studies and Forum Figures
Most of what is said about how AOD 9604 works comes from rodents and rabbits. In mice it cut weight gain and raised fat burning without the high blood sugar that full growth hormone caused. The animal studies used these amounts:
| Amount | Route | Length | Who | Source |
|---|---|---|---|---|
| 500 mcg per kg a day | By mouth | 19 days | Animals: obese Zucker rats | Ng and others, 2000 |
| Not given in the abstract | Slow-release mini-osmotic pump | 14 days | Animals: obese and lean mice | Heffernan and others, 2001, Int J Obes |
| Not given in the abstract | Into the belly cavity | 14 days | Animals: obese mice and mice without the beta-3 receptor | Heffernan and others, 2001, Endocrinology |
| 0.25 mg a week | Into the knee joint | Weekly shots, knees checked at 8 weeks | Animals: rabbits with damaged knee cartilage, 32 across four groups | Kwon and Park, 2015 |
Disclaimer: an animal amount never converts into a human dose, and no study ever tested the forum figure. Neither one is advice for you; a licensed clinician makes dose decisions.
A 2026 review from Poznan University of Medical Sciences gathered what bodybuilding forums post. It rates the human evidence for AOD 9604 as limited: short trials with weak body-fat signals and mostly lab work behind them. For AOD 9604 the forums post:
| Amount | Route | Length | Who | Source |
|---|---|---|---|---|
| 250 to 500 mcg a day | Under the skin, split into 2 to 3 shots, before exercise or in the morning before eating | 8 to 12 weeks | People: self-reported on bodybuilding forums | 2026 review |
Disclaimer: an animal amount never converts into a human dose, and no study ever tested the forum figure. Neither one is advice for you; a licensed clinician makes dose decisions.

How the AOD 9604 Math Works
Three short steps turn a vial into syringe units. You can check each one by hand.
strength (mcg per mL) = vial mg x 1,000 / water mL
volume (mL) = amount in mcg / strength
units = volume x 100
Worked through for a 5 mg vial with 2.5 mL of water and 200 mcg:
- 5 x 1,000 / 2.5 = 2,000 mcg per mL, so one unit holds 20 mcg.
- 200 / 2,000 = 0.1 mL.
- 0.1 x 100 = 10 units, and the vial holds 5,000 / 200 = 25 such draws.
Disclaimer: these round numbers show the steps only and are not an amount to use.

Picking the Water Volume
The water does not change how much AOD 9604 is in the vial. It changes how much each syringe mark holds. This table shows mcg per unit for three example vial sizes:
| Vial | 1 mL | 2 mL | 2.5 mL | 3 mL | 5 mL |
|---|---|---|---|---|---|
| 2 mg | 20 | 10 | 8 | 6.67 | 4 |
| 5 mg | 50 | 25 | 20 | 16.67 | 10 |
| 10 mg | 100 | 50 | 40 | 33.33 | 20 |
Disclaimer: these are per-unit strengths for sample vials, not amounts to draw.Fewer mcg per unit spreads a draw over more marks, which is easier to read. Too much water can push a draw past the barrel, so the calculator checks that too. For mL to units on any barrel, see our mL to units guide.

What the Trials and the FDA Say About Safety
The 2013 safety paper reports that headache was the most common complaint in the six trials, and it showed up on placebo too. At the highest capsule dose in table A there were more headaches, more diarrhea and more gas. The paper reports no change in IGF-1 and no change in glucose tests, and no antibodies to AOD 9604 were found.
Serious events were rare but not absent. In the single-dose capsule trial, one case of diarrhea at that top dose was rated possibly related. In the 12-week trial, five people on AOD 9604 had a serious event: four cancers, three of them skin cancers, and one fatty lump. The investigator judged none of the five related to the drug.
The FDA lists AOD-9604 on its April 2026 page of compounding ingredients that may pose serious risks, in the section for nominations that were withdrawn. Its note says there is little or no safety information, a risk of immune reactions, and serious adverse events that may be linked to AOD-9604, though the cause is not clear.
Some pages say the FDA gave AOD 9604 GRAS status, the term for food ingredients judged safe. We searched the FDA's GRAS notice inventory, which covers notices since 1998, and found no notice for AOD9604.
It is also banned in sport. A 2015 anti-doping lab paper says the World Anti-Doping Agency prohibits it, and that it had turned up in vials seized in the USA. Our guide to peptides to lose weight sets it beside the approved weight loss medicines.

How to Reconstitute an AOD 9604 Vial
A freeze-dried vial has to be dissolved before it can be measured. The steps below are general handling, not medical advice.
- Check the vial size. Find the mg printed on the AOD 9604 label. The examples on this page use a 10 mg vial.
- Swab the rubber tops. Clean the peptide vial and the water bottle with an alcohol pad and wait until both are dry.
- Draw the water. Fill a clean syringe with the amount of bacteriostatic water you settled on.
- Add it down the side. Push the water in slowly against the glass so it does not foam on the powder.
- Swirl until clear. Move the vial in small circles. Shaking can foam a peptide, so keep it gentle.
- Label the strength. Write the date and the mg per mL on the vial, and keep it as the supplier sheet directs.
Bacteriostatic water carries benzyl alcohol as a preservative. Its label says the bottle can be entered more than once and is for mixing or dissolving injectable drugs.
AOD 9604 Calculator Questions
How many units is 300 mcg of AOD 9604?
It depends on the water. In a 10 mg vial mixed with 2 mL, each unit holds 50 mcg, so 300 mcg is 6 units, or 0.06 mL. Mix the same vial with 5 mL and each unit holds 20 mcg, so 300 mcg becomes 15 units. Disclaimer: this is unit math only, not a dose.
How much bacteriostatic water should go in a 10 mg AOD 9604 vial?
No label sets a volume, because AOD 9604 has no approved product. The water only sets the strength. With 10 mg, 1 mL makes each unit hold 100 mcg, 2 mL makes it 50 mcg, and 5 mL makes it 20 mcg. Small amounts read more easily when each unit holds less.
Did the human trials inject AOD 9604 under the skin?
No. Two of the six trials gave single doses as a drip into a vein, measured per kg of body weight. The other four gave capsules or tablets by mouth, in mg. None of them tested a shot under the skin.
Does AOD 9604 raise IGF-1?
Not in the trials. The 2013 safety paper reports no change in IGF-1 in any of the six studies, and no change in glucose tolerance tests. That paper was written by people linked to the company that paid for the trials.
Is AOD 9604 approved by the FDA?
No. It is not an approved medicine, and a 2026 review says no major regulator has approved it for any use. On the FDA page of compounding ingredients that may be risky, it appears in the part for withdrawn nominations. We also found no FDA GRAS notice for it.
Is AOD 9604 allowed in sport?
No. A 2015 paper from an anti-doping lab says the World Anti-Doping Agency bans it, and that it had been found in vials seized in the USA.
Can this calculator tell me how much AOD 9604 to take?
No. It only turns the numbers you enter into mL and syringe units. It cannot tell you whether to use AOD 9604, how much, or how often. Those are questions for a licensed clinician.
