Measured In Skin
Several of the studies here measured skin directly, where the marketing points. Oral collagen peptides were pooled in two meta-analyses that reported improved hydration and elasticity versus placebo, with wrinkles favoured in the larger-course pooling; the mechanism and product side sit on the collagen peptides guide. Palmitoyl pentapeptide was applied to the faces of 93 women aged 35 to 55 in a split-face trial and reduced fine lines and wrinkles against a plain moisturiser, judged by image analysis and by a blinded grader. Acetyl hexapeptide-8, the topical peptide sold as argireline, reduced wrinkle depth by up to 30 percent over 30 days in a topography study of healthy volunteers. Copper tripeptide is on this list as GHK-Cu and its cosmetic form topical GHK-Cu: after laser resurfacing, blinded evaluators found no difference between groups, while patients rated their own skin quality higher on the copper regimen. The Growth Hormone Axis
On the growth hormone axis, the trials measured biomarkers and function rather than lifespan. Tesamorelin, a stabilised analog of growth hormone releasing hormone, was given by daily injection for 20 weeks to 152 adults aged 55 to 87, of whom 66 had mild cognitive impairment. The trial reported a favourable effect on cognition and on executive function, a 7.4 percent fall in body fat and a 117 percent rise in IGF-1 that stayed inside the physiological range. CJC-1295, a longer-acting analog of the same hormone, raised growth hormone and IGF-1 for days after a single injection in healthy adults aged 21 to 61, with IGF-1 still above baseline up to 28 days after multiple doses. A Pineal Peptide And Melatonin
Epithalamin, a peptide preparation from the pineal gland, was given as a course to healthy elderly subjects, and their plasma melatonin was tracked across a day. During the dark period, melatonin rose in subjects whose pineal activity started low and tended to fall in those whose function was already normal, so the study describes a modulation rather than a single direction of effect. The synthetic tetrapeptide derived from this line of work is sold as epitalon. The abstract does not state the sample size, which is part of why this row sits apart from the randomised skin and hormone trials above.

A hydration figure pooled from 1,721 patients is a different kind of evidence than a satisfaction score from 13 people after laser treatment, and a protein rise measured in a dish of skin explants is not a measured change on anyone's face. Do oral collagen peptides actually change skin, or just add water to it?+
Two meta-analyses pooled the randomised trials. The 2021 de Miranda review (PMID 33742704) pooled 19 trials and 1,125 participants and reported favourable hydration, elasticity and wrinkle results over courses around 90 days, with hydration and elasticity confirmed in a subgroup analysis. The 2023 Pu review (PMID 37432180) pooled 26 trials and 1,721 patients and reported improved hydration and elasticity. Both numbers are averages across trials of differing size and quality.
What is the difference between the topical peptides and the injected ones listed here?+
Route and endpoint. Palmitoyl pentapeptide, acetyl hexapeptide-8 and copper tripeptide were applied to skin and measured in skin, by wrinkle imaging and blinded grading. Tesamorelin and CJC-1295 were injected and measured in blood, in body composition and, for tesamorelin, in cognition. A result in facial skin is not a result in blood hormones, and neither is interchangeable with the other.
Do the growth hormone peptides here reverse aging?+
The cited trials measured specific outcomes over defined windows, not lifespan. The tesamorelin trial (PMID 22869065) measured cognition, body fat and IGF-1 over 20 weeks in adults aged 55 to 87 and reported a favourable cognition effect and lower body fat. The CJC-1295 trial (PMID 16352683) measured growth hormone and IGF-1 in healthy adults over four to seven weeks. Both are biomarker and function readouts, and both are prescription decisions made with a doctor.
Why does copper tripeptide appear more than once with different results?+
Because a row is a study. The post-laser trial (PMID 16847171) measured blinded wrinkle and erythema outcomes in 13 patients and found no difference between groups, while patient-rated skin quality favoured the copper regimen. The ex-vivo study (PMID 31603269) measured pro-collagen 1 and elastin secreted by human skin explants in culture. One reports a clinical outcome, the other a laboratory measurement, and collapsing them into a single verdict loses that.
